New England Journal of Medicine

Results from the phase 3 INDIGO trial, published in the New England Journal of Medicine, demonstrated that obexelimab significantly reduces the risk of disease flares in patients with IgG4-related disease.
IgG4-related disease is a chronic fibroinflammatory condition that can affect multiple organs, including the pancreas, kidneys, and glands. In this trial, 194 patients were randomly assigned to receive either obexelimab or placebo and were followed for 52 weeks.
The results showed a 56% reduction in the risk of disease flare requiring rescue therapy in the obexelimab group compared with placebo. Complete remission at week 52 was achieved in 37.1% of patients receiving obexelimab versus 19.6% in the placebo group. Obexelimab treatment also reduced the cumulative need for glucocorticoid rescue therapy.
These findings highlight the potential of targeted B-cell inhibition as a promising therapeutic approach for autoimmune and inflammatory diseases, while potentially reducing dependence on glucocorticoid treatment.
